Your first Pathogenic game run should almost always be Bacteria. It is unlocked from the menu, teaches ranged and melee grafting without exotic slot rules, and its plasmid tree includes straightforward health and organelle unlocks that make the second attempt much easier.

Bacteria — the teaching pathogen

Bacteria’s body is compact enough to read in the organelle editor on a first run. You can attach a secretor for range, a spike for melee defense, and one mitochondria without fighting awkward slot angles. Most community “first clear” routes assume Bacteria plasmids that add a stash slot and one extra mutation choice.

Helminth and Spore — opposite combat poles

Helminth wants you in the immune cell’s face: spikes delete projectiles and knock enemies back, which pairs with phagocytosis challenge mode later. Spore instead floods the room with minions; its power spikes when mitochondria overcharge spawn rates. If you dislike positioning, start Spore only after you understand flagella pacing.

Amoeba and Nanobot — body shape specialists

Amoeba rewards dragging organelles around freely — repositioning a secretor behind a spike changes reach and mass in ways other pathogens cannot. Nanobot is the opposite: stack armor cores, absorb mistakes, and win boss DPS checks through sustained overcharge rather than dodging every bullet.

Protozoa and Virus — unlock-tier carry picks

Protozoa becomes one of the strongest speed runners once its dash plasmids are online; until then it feels fragile in Lungs airflow rooms. Virus is the secretor playground — infusers chain farther on its frame, but you need prior clears to unlock it and feed its tree.

How does each plasmid tree differ?

Bosses drop plasmid fragments spent only on the pathogen you played that run. Trees are not shared — Bacteria health nodes do nothing for Virus until you grind Bacteria again. Plan one “main” species for metaprogression each week.

Fragments are pathogen-locked

Starting unlocks inside the tree

Several pathogens hide alternate starting organelles behind plasmid nodes — not just stat bumps. Check the codex after each purchase; some nodes swap your default weapon entirely, which changes early-room tempo more than a raw +10% damage line.

Once a tree row is funded, co-op or Diatom runs become the fastest way to test new nodes without risking solo streaks — covered next.

What about Diatom co-op?

Diatom is not an eighth roster entry in the table above — it is a co-op pathogen where two halves share one host body (or one player controls both halves in solo Diatom mode since the July 22 patch). Pick it from the same select screen after enabling co-op; press X on Diatom to queue solo split-control if you have no partner.

Unlock achievements and most plasmid fragment farms still route through the seven solo pathogens. Use Diatom for practice and fun duos, not for your first account-wide unlock push.
