Pathogenic game builds are not a single loadout — they are live organelle graphs you reshape every evolution screen. On Steam you graft flagella, mitochondria, secretors, and spikes onto one of 7 pathogens, chain overcharge through connected slots, and push through 6 organ biomes until the immune system breaks. This page covers the meta build archetypes, mitochondria wiring rules, pathogen fit, and co-op role splits for Pathogenic — not biology textbook pathogen definitions.
Pair with the organelles tier list for ranked graft picks, the pathogens hub for starter kits, and the plasmid tree for between-run power. Verified July 27, 2026 · Aberrant Labs · Slug Disco · Steam app 3808690.

| Build archetype | Core organelles | Win condition | Best pathogen fit |
|---|---|---|---|
| Lacerator Tendril · Extensor · Resonant Cavity | Zero stamina swing spam at boss range | Amoeba · shape-shift blob | |
| Laser Emitter · Homing internal · metabolic mitochondria | Homing beams scale into screen wipe | Helminth · Nanobot | |
| Metabolic Mitochondria · overcharge supports | Pickup loops feed overcharge health and DNA | Any pathogen with center-line plasmids | |
| Bacteriophage Launcher · swarm spawners | Minions absorb aggro; you stack buff auras | Bacterium · Diatom co-op | |
| Pyroflagellum · Gyrase Wheel · burn secretors | Orbitals + ignite stacks melt waves | Flagella-heavy starters | |
| Spike shells · tail reflect passives | Soft-body collisions + reflected bullets | Helminth tail builds |
Every pathogenic game build starts at the evolution screen: defeat immune cells, pick a new organelle or mutation, then rotate and reposition grafts before you lock the choice. Community footage (JammyGiraffe tips, SlayerAnims beginner breakdown) repeats the same lesson — you can preview layouts, drag internal and external slots, and cancel out without committing until the network looks right.
Build power comes from three layers: base organelle stats, connection networks (supports buffing weapons), and mitochondria overcharge feeding the chain. Raw damage numbers on a lone secretor lie; a mediocre emitter wired through two supports and a triggering mitochondria often beats a “S-tier” graft floating unconnected.
Between organs you also spend DNA on run mutations and cores at shops. Builds that ignore economy — never picking up DNA or recycling Ephemeral grafts — stall even when DPS looks fine. See the beginner guide for currency basics and the walkthrough for when to push the next biome.

Two organelles can overlap on your soft-body mesh and still not share bonuses. Valid links show flow arrows that change color when powered; hover any piece to see what it receives from neighbors. If your mitochondria needs ten successive hits to overcharge, it must sit on the same network as a weapon that actually lands those hits — not a mobility flagellum on the other side of the cell.
Each pathogen exposes internal, external, and core attachment points with connection caps — commonly up to three links on a single node (Amoeba melee guides stress this when placing Conductive Core blobs). Filling a hub slot early blocks later synergies; plan your spine before you graft luxury pieces.
Cancerous grafts spike stats but transfer on clone. Ephemeral pieces break yet recycle into resources — great for economy builds, terrible if you anchored your only overcharge path on them. Read tags before you evolve; the organelles database lists trait behavior.
Meta pathogenic game builds cluster into a handful of archetypes. Names vary in Discord and YouTube titles — “BIG BURN”, “orbital death”, “minion brain delete” — but they all map back to organelle families. Use the table as a route picker; exact graft names shift with patches, so cross-check the organelles tier list.

| Archetype | Tier ceiling | Strength | Weakness |
|---|---|---|---|
| S | Insane single-target DPS when Resonant Cavity stacks attack speed | Needs Extensor range or you eat boss hitboxes | |
| S | Homing + laser pairs scale into boss melters | Weak until homing internal comes online | |
| A | Pickup and recycle loops fuel overcharge sustain | Slow burst until networks finish | |
| A | Swarm clears waves while you reposition | Struggles vs anti-swarm macrophage packs | |
| A | Area denial in lung/heart corridors | Needs flagella mobility to kite hazards | |
| B | Tail reflect and spike shells trivialize bullet rooms | Brain boss patterns still demand burst |
MarvGoneRogue’s Amoeba brain bully runs show the template: Lacerator Tendril on epic, Extensor for swing reach, Resonant Cavity overriding attack rate (community tests report 150%+ fire rates when hits connect). Pair with plasmid nodes that remove stamina tax on swings. You play inside boss hitboxes — soft-body physics become your dodge — so read controls for dash/sprint stamina.
The “broken combo” clips pair Laser Emitter with a homing internal organelle, then stack Blast Assist Mortar or rocket secretors for delete-key boss phases. Metabolic Mitochondria that grant overcharge health on pickups turn self-damage mortars into sustain engines. Helminth’s tail passives (reflect, dodge explosions) add spice but are optional — the core is homing + multiplier secretors.
Energy builds treat mitochondria as the spine, not a stat stick. Pickup-triggered overcharge, recycle bonuses, and DNA-on-collection effects snowball when you clear rooms without missing drops. They start slower than spike or laser spikes but stabilize long heart and brain routes where raw burst alone falls off.
Bacteriophage Launcher and swarm-style secretors turn you into a bullet-heaven commander. Minions hold aggro while you layer auras. Haelian and MarvGoneRogue brain runs prove the ceiling — but until spawn rate items appear, treat this as a mid-run pivot, not Act 1 pick.
Orbitals plus Pyroflagellum or burn infusions excel in wide lung and heart chambers. You kite using flagella, let orbitals and ignite stacks clear density, then burst bosses with a secondary secretor. Update 4 added Tri-Phase Tendril and Gyrase Wheel as melee-adjacent options if you want hybrid tech.
Mitochondria are the power plant of every serious pathogenic game build. Overcharge is not “more damage” generically — each mitochondria specifies how it activates: successive hits, pickups, on-damage procs, or timed ticks. Feed that trigger correctly or the rest of the network idles.

A mitochondria that needs 10 successive attacks belongs beside a fast secretor or melee tendril — not a slow mortar. Pickup-based mitochondria want routes through core-heavy rooms and shop recycling. On-damage cores fit tanky spike builds that intentionally collide. Match trigger to weapon cadence before you judge an organelle “bad”.
Supports buff weapons they touch if mitochondria power reaches them first. A support buffing a homing laser can raise projectile count, ailment chance, or overcharge spill — but only when arrows show active flow. Re-run the evolution preview after every new piece; five seconds beats a twenty-minute doomed organ.
Between runs, plasmid nodes like Conductive Core (Amoeba center-line growth chance) or global damage/stamina perks define which archetypes are honest on your account. A spike build without stamina plasmids still works — it just arrives later. Plan metaprogression on the plasmid tree page before you force a meme build on run one.
Pathogens are build envelopes — starting slots, movement quirks, and exclusive plasmid branches. You can force any archetype on any character, but friction varies. Use the pathogens hub for full kits; below is the meta shorthand for pathogenic game builds on Steam.
| Pathogen | Natural archetype | Plasmid note | Avoid if… |
|---|---|---|---|
| Amoeba | Melee spike / flexible slots | Conductive Core center-line growth | You refuse close-range boss play |
| Helminth | Secretor / tail reflect | Tail reflect & dodge explosion passives | You need strafe-heavy kiting |
| Bacterium | Minion / swarm | DNA discount leveling | You want solo laser burst only |
| Diatom | Co-op dual control | Split-body mechanics | Solo only — control overhead |
| Nanobot | Tech / secret unlock routes | Evolution thresholds for hidden pathogen | You want zero setup time |
Needs 20% less DNA to level per community testing, shape-shifts freely, and loves Lacerator Tendril setups. Internal/external freedom is the selling point — but connection caps punish greedy hub placement. Best first pathogen for learning evolution screen craft.
Brown kit, tail blocks shots, mouse-move recommended. Tail reflect and contact-damage passives enable reflect memes, but the mainstream route is mortar + homing laser scaling. Cannot strafe like Amoeba; positioning is deliberate.
Diatom shines in co-op: one player anchors flagella mobility, the other stacks secretors. Solo Diatom is viable with dual-stick practice but not where most build guides start.
Local split-screen pathogenic game co-op rewards complementary builds, not duplicate DPS. One pathogen stacks flagella and spike shells to body-block macrophages; the other wires secretors and mitochondria for burst. Shared room clears mean pickup economy benefits both — coordinate who takes evolution picks for network pieces vs. luxury damage grafts.

Tank role: spikes, armor cores after boss nodes, movement flagella. Carry role: homing lasers, Resonant Cavity, metabolic mitochondria. Swap evolution picks when the carry’s network is complete and the tank still needs slot coverage.
Most failed pathogenic game builds share the same errors — fixable without rerolling the pathogen.
Locking the first organelle offered without rotating loses free DPS. Treat every evolution as a mini puzzle; cancel until arrows flow.
Fighting momentum instead of surfing it wastes stamina. Lateral dashes and wall ricochets beat straight retreats into macrophages — the gelatinous body is a weapon, not a handicap.
Stacking five secretors with zero mitochondria or mobility dies to lung rush packs. Every archetype needs one movement piece, one power spine, one burst win condition.
Cancerous transfers on clone can gut your main weapon if the wrong piece duplicates. Clone deliberately or skip until you understand trait rules on the organelles page.

Community footage favors homing laser + metabolic mitochondria and Amoeba melee tendril stacks for brain boss DPS. Patches move exact graft names — use the organelles tier list for current ranks.
Core networks yes, but split roles — one tank mobility/spikes, one secretor burst. See the co-op guide.
